目的 :研究雾化吸入γ 干扰素对免疫低下大鼠肺部抗感染能力的增强作用。方法 :醋酸考的松皮下注射复制大鼠免疫低下模型 ,检测气管内注射白色念珠菌或气管内注射白色念珠菌并雾化吸入γ 干扰素 1、3、7d的免疫低下大鼠肺泡巨噬细胞的吞噬及杀菌功能 ,抗原提呈功能 ,以及培养上清中TNF α活性 ,并于第 7天进行左肺白色念珠菌培养计数。结果 :雾化吸入γ 干扰素组大鼠肺泡白色念珠菌计数显著少于免疫低下组大鼠 (P <0 0 5 )。雾化吸入γ 干扰素组大鼠肺泡巨噬细胞的吞噬及杀菌百分率显著高于免疫低下组大鼠 (P <0 0 1)。雾化吸入γ 干扰素组大鼠肺泡巨噬细胞Ia抗原表达的阳性率显著高于免疫低下组大鼠(P <0 0 1)。雾化吸入γ 干扰素组大鼠肺泡巨噬细胞培养上清中TNF α的活性显著高于免疫低下组大鼠(P <0 0 5 )。结论 :雾化吸入γ 干扰素可明显增强免疫低下大鼠肺部抗白色念珠菌感染能力。
OBJECTIVE: To study the enhancing effect of inhaled interferon gamma (IFN-γ) on anti-infection ability of lung in immunocompromised rats. METHODS: Rat model of immunosuppression was established by subcutaneous injection of test-cortisone acetate. The immunocompromised alveolar macrophages of mice with Candida albicans intratracheal instillation or Candida albicans intratracheal intratracheal intratracheal instillation of IFN- Phagocytosis and bactericidal function, antigen presenting function, and TNFα activity in the culture supernatant, and Candida albicans culture count was performed on the 7th day. Results: Candida albicans counts in alveolar group were significantly less than those in immunosuppressed group (P <0.05). The percentage of phagocytic and bactericidal percentage of alveolar macrophages in inhaled IFN-γ group was significantly higher than that in immunocompromised group (P <0.01). The positive rate of Ia antigen expression in alveolar macrophages of inhaled IFN-γ group was significantly higher than that in immunocompromised group (P <0.01). The activity of TNFα in alveolar macrophage culture supernatant of inhaled IFNγ group was significantly higher than that in immunocompromised group (P <0.05). CONCLUSION: Inhaled interferon gamma can significantly enhance the anti-Candida albicans infection in the lungs of immunocompromised rats.
