目的:比较在4种无硫化产地加工条件下,浙贝母的产品外观及内在质量,为筛选出技术先进、适合推广的浙贝母产地加工工艺提供参考。方法:以成品外观、含水量、贝母素甲和贝母素乙含量为指标,比较贝壳粉吸附、生切烘干、冷冻干燥、微波干燥4种产地加工工艺对浙贝母外观和质量的影响。采用HPLC-ELSD测定贝母素甲和贝母素乙含量,流动相乙睛-水-二乙胺(70∶30∶0.03),流速0.8 m L·min-1,漂移管温度85℃,载气流速2.2 L·min-1。结果:各工艺条件下成品外观差异较大,以冷冻干燥品为最佳;含水量均符合2010年版《中国药典》规定。贝母素甲和贝母素乙含量测定的线性范围分别为0.925~4.625,0.94~4.7μg;贝母素甲和贝母素乙总量的平均回收率98.58%。各工艺条件下贝母素甲和贝母素乙总量存在一定差异,冷冻干燥品中含量最高,微波干燥品和生切烘干制品次之,贝壳粉吸附制品含量最低。结论:浙贝母质量应从源头上注重产地加工的安全性,建议可采用微波干燥代替传统的产地加工方法。
OBJECTIVE: To compare the appearance and intrinsic quality of Fritillaria thunbergii in the processing conditions of four sulfur-free production areas, and to provide a reference for selecting the processing technology of the origin of Fritillaria thunbergii which is technologically advanced and suitable for popularization. Methods: According to the appearance, water content, Fritillaria a and Peiminine B content, the appearance and quality of Fritillaria ussuriensis were compared with the processing methods of shellfish powder adsorption, raw cut drying, freeze drying and microwave drying influences. HPLC-ELSD was used to determine the contents of Fritillariae and Peiminine. The mobile phase was acetonitrile-water-diethylamine (70:30:0.03), the flow rate was 0.8 m L · min-1, the temperature of the drift tube was 85 ℃, Gas flow rate 2.2 L · min-1. Results: The appearance of the finished products varied greatly under different process conditions, and the freeze-dried products were the best. The water content accorded with the 2010 edition of “Chinese Pharmacopoeia”. The linear ranges of Fritillaria and Fritillaria were 0.925-4.625 and 0.94-4.7μg, respectively. The average recoveries of Fritillaria and Fritillaria were 98.58%. There was a certain difference in the total amount of Fritillariae and Peiminidil under various processing conditions. The content of freeze-dried product was the highest, followed by the microwave-dried product and raw-cut product, and the content of shellfish powder-adsorbed product was the lowest. Conclusion: The quality of Fritillaria cirrhosa should pay attention to the safety of origin processing from the source. It is suggested that microwave drying should be used instead of the traditional processing method of origin.
