目的了解小鼠感染伯氏疟原虫后Toll样受体(TLRs)的免疫作用。方法用伯氏疟原虫感染小鼠,采用RT-PCR、ELISA等方法测定TLRs的mRNA和下游炎症因子血清水平。结果小鼠感染伯氏疟原虫后TLR2、9、11的mRNA分别升高3、3.5和6倍;在感染后96h,IL-6、TNF-α及IL-12均达到峰值,分别为10 023U/ml、485pg/ml和186pg/ml;IL-18持续升高,在144h为4 225pg/ml。结论小鼠感染伯氏疟原虫后TLRs mRNA表达上调,并增强下游炎症因子的表达。
Objective To understand the immune effects of Toll-like receptors (TLRs) in mice infected with Plasmodium berghei. Methods Mice were infected with Plasmodium berghei and RT-PCR, ELISA and other methods were used to determine the mRNA level of TLRs and the serum levels of downstream inflammatory cytokines. Results The mRNA levels of TLR2, 9 and 11 in mice were increased by 3, 3.5 and 6 folds respectively after infection with P. berghei. After 96 h, the levels of IL-6, TNF-α and IL-12 peaked at 10 023U / ml, 485 pg / ml, and 186 pg / ml; IL-18 persistently increased at 4 225 pg / ml at 144 h. Conclusion Mice infected with Plasmodium berghei TLRs mRNA expression increased, and enhance the expression of downstream inflammatory cytokines.
