目的分离3起食物中毒患者粪便及食物加工工具样本中的病原菌,对分离菌株进行表型和毒力基因鉴定。方法可疑病原菌的分离与鉴定按照国标GB/T4789-2003和GB/T4789.7-2008规定的方法进行;用PCR方法对检出的副溶血弧菌(Vibrio parahaemolyticus,Vp)进行Vp种属特异基因(320bp)和毒力基因tdh(耐热溶血素434bp)、trh(耐热相关溶血素250bp)检测;用环介导恒温扩增(LAMP)方法对分离株tlh(不耐热溶血素229bp)基因进行检测。结果从3起食物中毒的患者、厨师粪便和食品加工环节共27件样品中检出7株Vp,其中加工环节熟食冷菜间刀具涂抹物中检出1株,患者粪便中检出6株。3起食物中毒7株菌血清型分属为O10∶K66、O2∶K3、O1∶K33,每起食物中毒分离株的生物学性状、血清型和药敏试验结果均一致。7株Vp均产生神奈川现象(KP)、出现相同Vp种属特异基因条带、均检出tlh毒力基因,但tdh、trh基因则检测为阴性。结论3起食物中毒均为tlh+/tdh-/trh-不同血清型Vp污染引起。
OBJECTIVE To isolate three pathogenic bacteria in food poisoning cases from feces and food processing tools, and to identify the phenotype and virulence genes of the isolated strains. Methods Isolation and identification of suspected pathogens were carried out according to the methods specified in GB / T4789-2003 and GB / T4789.7-2008. Vp genus-specific genes (Vp) were detected by PCR in Vibrio parahaemolyticus (Vp) (320bp) and virulence genes tdh (heat-resistant hemolysin 434bp) and trh (heat-related hemolysin 250bp). The isolates tlh (heat-labile hemolysin 229bp) were detected by ring-mediated isothermal amplification (LAMP) Gene testing. Results Seven strains of Vp were detected in 27 samples of food poisoning patients, cook faeces and food processing sectors. Among them, one Vp was detected in the tool smears of cooked food and six were detected in the feces of the patients. 3 strains of food poisoning 7 strains belonging to the serotype O10: K66, O2: K3, O1: K33, each of the biological characteristics of food poisoning isolates, serotypes and susceptibility test results are the same. 7 strains of Vp produced Kanagawa phenomenon (KP), the same Vp species gene-specific gene bands were detected, were detected tlh virulence genes, but tdh, trh gene was tested as negative. Conclusion The three food poisoning are caused by Vp contamination of different serotypes of tlh + / tdh- / trh-.
