目的探讨p53和MDM2在胃腺癌组织中的表达及临床意义。方法采用免疫组织化学染色方法检测94例胃腺癌组织中p53和MDM2的表达水平,分析两者表达的相关性。结果 p53(突变型)的检出率为36%(34/94),在这些可检出突变型p53蛋白表达的胃腺癌原发灶组织中并非所有癌细胞均为p53阳性表达,其阳性率差异较大,从10%99%,均数约30%;在62.8%(59/94)的胃腺癌原发灶组织中可检测到MDM2蛋白表达,定位于胞质和胞核,呈异质性表达;MDM2蛋白表达水平在p53野生型胃腺癌组织(免疫组织化学染色阴性)中高于p53突变型胃腺癌组织(免疫组织化学染色阳性)。结论在胃腺癌组织中,p53和MDM2呈异质性表达;在部分胃腺癌细胞中,高水平MDM2蛋白表达是限制野生型p53蛋白积聚活化的主要原因之一。
Objective To investigate the expression and clinical significance of p53 and MDM2 in gastric adenocarcinoma. Methods The expression of p53 and MDM2 in 94 cases of gastric adenocarcinoma was detected by immunohistochemical staining and the correlation between them was analyzed. Results The positive rate of p53 (mutant type) was 36% (34/94). Not all of the cancer cells in the primary lesions of gastric adenocarcinoma with detectable mutant p53 protein expression were positive for p53. MDM2 protein expression was detected in 62.8% (59/94) of primary gastric adenocarcinoma tissues and localized in the cytoplasm and nucleus The expression of MDM2 protein was higher in p53 wild type gastric adenocarcinoma tissues (immunohistochemical staining negative) than in p53 mutant gastric adenocarcinoma tissues (immunohistochemical staining). Conclusions The expression of p53 and MDM2 is heterogeneous in gastric adenocarcinoma. In some gastric adenocarcinoma cells, the high level of MDM2 protein expression is one of the main reasons that limit the activation of wild-type p53 protein.
