目的:探讨补肾壮骨颗粒含药血清对去卵巢骨质疏松大鼠骨髓间充质干细胞(MSCs)体外增殖的影响。方法:选用10只6月龄SD大鼠去卵巢制作骨质疏松模型,对骨质疏松骨髓间充质干细胞进行分离和体外培养,传代后的MSCs分为OP组、OP+补肾壮骨颗粒含药血清组和OP+空白血清组,倒置显微镜观察各组MSCs的生长情况,进行形态学观察,并用四甲基偶氮唑蓝(MTT)法测定生长曲线。结果:OP+含药血清组,MSCs呈典型的集落生长,细胞呈长梭形,突起细长且多,细胞增殖较旺盛,细胞数量不断增加,随着传代次数的增多,细胞增殖速度减慢。OP组及OP+空白血清组MSCs接种后胞体立体感下降,体积较大,细胞偏圆形,突起收缩,增殖速度较慢,细胞数量减少;随着传代次数的增多,细胞增殖速度显著减慢。OP+补肾壮骨颗粒含药血清组骨髓间充质干细胞增殖能力明显提高,生长曲线有显著性差异。结论:补肾壮骨颗粒含药血清能促进去卵巢骨质疏松大鼠MSCs的增殖。
Objective: To investigate the effect of Bushen Zhuanggu granule-containing serum on proliferation of bone marrow mesenchymal stem cells (MSCs) in ovariectomized rats in vitro. Methods: Osteoporosis models were made in 10 6-month-old SD rats by ovariectomy. Osteoporotic bone marrow mesenchymal stem cells were isolated and cultured in vitro. The MSCs were divided into OP group, OP + Bushen Zhuanggu granules Serum group and OP + blank serum group. The growth of MSCs in each group was observed by inverted microscope, and the morphology was observed. The growth curve was determined by MTT assay. Results: In the OP + drug-containing serum group, MSCs showed the typical colony growth. The cells were long fusiform with protuberant slender and many cells proliferating. The number of cells increased continuously. With the increase of passage times, the proliferation of MSCs was slowed down. After inoculation, MSCs in OP group and OP + blank serum group showed decreased stereopsis, larger volume, rounded cells, contracted protrusions, slowed down proliferation and reduced the number of cells. With the increase of passage times, the proliferation rate of MSCs decreased significantly. The proliferation of bone marrow mesenchymal stem cells of OP + Bushen Zhuanggu granule-containing serum group was significantly improved, the growth curve was significantly different. Conclusion: Bushen Zhuanggu Granule-containing serum can promote the proliferation of MSCs in ovariectomized rats.
