[目的]调查正常人群中脑膜炎奈瑟氏菌携带率,并进行方法评价。[方法]采用实时荧光PCR法检测不同地区不同年龄组人群鼻咽拭子中NmDNA,并与传统的培养方法进行比较评价。[结果]正常人群999份鼻咽拭子中NmDNA检出率为8.41%,嵌合荧光PCR法方法的灵敏度、特异性分别为:100%,100%,阳性预测值(PPV)为13.09%,阴性预测值(NPV)为100%,检出限为250cfu/m,荧光PCR方法检出的时间为0.5d。而培养法,阳性结果需要3~4d,阴性结果2d。[结论]嵌合荧光定量PCR法具有灵敏、特异的特点,特别适合正常人群的带菌率调查,也可用于疾病的快速诊断。
[Objective] To investigate the carrier rate of Neisseria meningitidis in normal population and to evaluate the method. [Methods] Real-time fluorescent PCR was used to detect nasopharyngeal swabs NmDNA in different age groups and compared with the traditional culture methods. [Results] The detection rate of NmDNA in the nasopharyngeal swab of 999 normal subjects was 8.41%. The sensitivity and specificity of the method for chimeric fluorescence PCR were 100%, 100%, 13.09%, respectively. [Conclusion] The method of chimeric fluorescence quantitative PCR has the characteristics of sensitivity and specificity, and is especially suitable for the investigation of carrier rate in normal population and also for the rapid diagnosis of diseases.
