目的:研究三乙酰白藜芦醇(TRES)对胰腺癌Panc-1细胞增殖及STAT3、NF-κB的影响。方法:采用MTT法检测Panc-1细胞增殖活性,克隆形成、细胞划痕实验分别检测Panc-1细胞的分化和迁移侵袭能力,以及Western blotting法检测STAT3和NF-κB的蛋白表达。结果:MTT实验结果表明TRES可以抑制Panc-1细胞的增殖。与对照组比较,克隆形成和细胞划痕实验显示TRES抑制了Panc-1细胞的分化和迁移侵袭能力(P<0.05),Western blotting结果显示TRES下调了磷酸化的STAT3和NF-κB蛋白表达,表明TRES抑制了STAT3和NF-κB的活化。结论:TRES可以抑制Panc-1细胞增殖,可能具有抗肿瘤活性。
Objective: To investigate the effect of TRES on the proliferation, STAT3 and NF-κB in pancreatic cancer Panc-1 cells. Methods: MTT assay was used to detect the proliferation and migration of Panc-1 cells. Clonal formation and cell scratch assay were used to detect the differentiation and migration of Panc-1 cells. Western blotting was used to detect the protein expression of STAT3 and NF-κB. Results: MTT results showed that TRES could inhibit the proliferation of Panc-1 cells. Compared with the control group, TRES inhibited the differentiation and migration of Panc-1 cells (P <0.05). Western blotting showed that TRES downregulated phosphorylated STAT3 and NF-κB protein expression, TRES inhibited the activation of STAT3 and NF-κB. Conclusion: TRES can inhibit the proliferation of Panc-1 cells and may have anti-tumor activity.
