目的研究大黄素对脂多糖激活后巨噬细胞高迁移率族蛋白B1(HMGB1)释放的影响及其机理。方法通过细胞培养和动物实验,观察大黄素对脂多糖激活后巨噬细胞株RAW264.7HMGB1释放、HMGB1mRNA表达和HMGB1核浆转移的作用,以及对内毒素血症小鼠血清HMGB1水平和存活率的影响。结果大黄素明显抑制脂多糖激活后巨噬细胞HMGB1的基因表达、核浆转移和胞外释放,降低内毒素血症小鼠血清HMGB1水平并提高存活率。结论大黄素具有抑制HMGB1胞外释放作用。
Objective To study the effect of emodin on the release of high mobility group box 1 (HMGB1) from macrophages after lipopolysaccharide activation and its mechanism. Methods The effects of emodin on macrophage RAW264.7 HMGB1 release, HMGB1 mRNA expression and HMGB1 nuclear plasma metastasis after lipopolysaccharide activation were observed by cell culture and animal experiments. The serum HMGB1 levels and survival rate in endotoxemia mice were also observed. influences. Results The emodin significantly inhibited the expression of HMGB1 macrophage, the plasmocytoplasmic transfer and extracellular release after lipopolysaccharide activation, reduced serum HMGB1 levels in endotoxemia mice and increased survival. Conclusion Emodin can inhibit the extracellular release of HMGB1.
