目的 :通过体外实验评价吲哚美辛对人喉癌Hep 2细胞系的抑制作用。 方法 :应用不同浓度的吲哚美辛作用于人喉癌Hep 2细胞系 ,进行细胞形态观察 ,锥虫蓝染色法检测细胞生长情况和流式细胞仪分析细胞DNA含量。Hep 2细胞经吲哚美辛作用后 ,撤药培养 ,绘制生长曲线 ,并用软琼脂集落形成实验检测肿瘤细胞非锚着生长能力 ,琼脂糖滴法检测肿瘤细胞移动能力 ,单层细胞侵袭实验计算肿瘤侵袭指数。结果 :Hep 2细胞系在吲哚美辛作用下 ,细胞生长形态改变 ,细胞增殖缓慢 ,流式细胞仪分析形成明显的亚二倍体峰。细胞毒性与药物剂量及作用时间有依赖关系。吲哚美辛作用后的Hep 2细胞软琼脂集落形成数目减少 ,细胞移动能力下降 ,单层细胞侵袭指数降低 ,但生长曲线较对照组无明显差异。结论 :吲哚美辛抑制人喉癌细胞系Hep 2生长 ,诱导凋亡 ,降低细胞的侵袭性。
OBJECTIVE: To evaluate the inhibitory effect of indomethacin on human laryngeal carcinoma Hep 2 cell line in vitro. Methods: Different concentration of indomethacin was applied to human laryngeal carcinoma Hep 2 cell line. Cell morphology was observed. Cell growth was detected by trypan blue staining and DNA content was analyzed by flow cytometry. After treated with indometacin, Hep 2 cells were withdrawn from the culture medium and the growth curve was drawn. The ability of non-anchored growth of tumor cells was detected by soft agar colony formation assay. The ability of tumor cells to move was detected by agarose dropping assay. Tumor invasion index. Results: Under the action of indometacin, Hep 2 cells changed morphology and slowed cell proliferation. The sub-diploid peak was formed by flow cytometry. Cytotoxicity and drug dose and the role of time-dependent. After indomethacin treatment, Hep 2 cells reduced the number of soft agar colonies, decreased the ability of cell migration, and decreased the invasive index of monolayer cells, but the growth curve had no significant difference compared with the control group. Conclusion: Indometacin inhibits the growth of human laryngeal carcinoma cell line Hep 2, induces apoptosis and reduces cell invasiveness.
