能迅速和准确地测定百日咳毒素(PT)含量的酶联免疫吸附测定(ELISA)对制造、纯化和检定百日咳菌苗有重要意义。本文建立和比较了两种测定百日咳毒素(PT)含量的ELISA法。在试验中制备了此两种夹心法所需要的多克隆兔抗百日咳杆菌抗体(RaBp)和鼠单克隆抗PT抗体(Mca PT)。被测定的材料为经纯化的或未经纯化的百日咳抽提液,其方法是依次用5%饱和度和含15mM磷酸钠的1M NaCl(pH8)浓缩和抽提
Enzyme-linked immunosorbent assays (ELISA), which allow rapid and accurate determination of pertussis toxin (PT) levels, are important for the manufacture, purification and characterization of pertussis vaccine. This article established and compared two ELISA method for the determination of pertussis toxin (PT) content. The polyclonal rabbit anti-pertussis antibody (RaBp) and the murine monoclonal anti-PT antibody (Mca PT) required for these two sandwich methods were prepared in experiments. The material tested was a purified or unpurified pertussis extract by concentration and extraction in succession with 5% saturation and 1 M NaCl (pH 8) containing 15 mM sodium phosphate
