H615系615纯系小鼠自发性肝细胞性肝癌,经埋块法移植传代成功,至今已20代,移植率在90%以上,潜伏期为15~35天,生长缓慢,随着肿瘤的生长,机体逐渐消瘦,出现恶病质,乃至死亡,在90天时死亡率达68.4%。瘤细胞分化较好,有小梁形成,但细胞学异型性明显。组织化学显示瘤细胞内核酸丰富,糖元缺失,酸性磷酸酶及β—葡萄糖醛酸酶分布不均匀。电镜观察与光镜所见基本符合,瘤细胞多数分化较高,细胞器与膜系统较完善,但由于瘤细胞处子增殖与退变的不同阶段,因而超微结构也表现为多形性。该肿瘤尚产生AFP,其血清与抗鼠AFP单向抗血清产生沉淀反应,用组织化学及生物化学方法证实瘤细胞内含有γ—谷氨酰转肽酶,可见H615含有肝细胞癌的两个主要肿瘤标志物。根据以上特点,该小鼠肝癌模型与人类肝细胞性肝癌极为相似,且其病程较长,可用子肝癌的基础理论研究及实验治疗。
H615 series 615 pure mouse spontaneous hepatocellular carcinoma was successfully transplanted by embedding method. It has been passed for 20 generations. The transplantation rate is over 90%, and the incubation period is 15-35 days. The growth is slow. With the growth of the tumor, The body gradually became thin, with cachexia, and even died. The mortality rate reached 68.4% at 90 days. The tumor cells were well differentiated and formed trabeculae, but cytological atypicality was evident. Histochemistry shows that the tumor cells are rich in nucleic acids, lack of glycogen, and the distribution of acid phosphatase and β-glucuronidase is not uniform. Electron microscopy observations were basically consistent with those seen with light microscopy. Most of the tumor cells were highly differentiated and the organelles and membrane system were relatively complete. However, due to the different stages of tumor cell proliferation and degeneration, the ultrastructure also showed polymorphism. The tumor still produces AFP, its serum reacts with anti-rat AFP unidirectional antiserum, and histochemical and biochemical methods confirm that the tumor cells contain gamma-glutamyl transpeptidase, showing that H615 contains two hepatocellular carcinomas The main tumor marker. Based on the above characteristics, the mouse liver cancer model is very similar to human hepatocellular carcinoma, and its long course of disease can be used for basic theoretical research and experimental treatment of sub-hepatoma.
