目的:研究Leydig细胞在抗感染中的免疫调节作用。方法:以溶脲脲原体(UU)直接注入大鼠膀胱拟上行性感染的途径,制备大鼠睾丸感染的动物模型,分别在UU感染的1周、2周、3周处死大鼠,分离取得睾丸组织,观察组织的病理变化,并分离获得高纯度的Leydig细胞,抽提总RNA,用RTPCR方法比较UU感染组与正常对照组之间IL1、IL6、TGFβ和Fas、FasLmRNA表达的差异。结果:与正常对照组相比,UU感染的大鼠睾丸组织发生明显的病理改变,IL1、IL6、TGFβ均升高,Fas表达下降、FasL表达升高。结论:大鼠Leydig细胞在抗感染免疫中,可通过改变IL1、IL6、TGFβ和Fas、FasL转录表达格局来发挥抗UU的免疫调节作用。
Objective: To study the immunomodulatory effect of Leydig cells in anti-infective. Methods: The animal model of testicular infection in rats was prepared by direct injection of UU (Ureaplasma urealyticum) into the aspirating pathological changes of the bladder in rats. The rats were sacrificed at 1 week, 2 weeks and 3 weeks after UU infection respectively. The pathological changes of the tissues were observed and the Leydig cells were isolated and purified. The total RNA was extracted and the differences of IL1, IL6, TGFβ, Fas, FasL mRNA expression between UU infection group and normal control group were compared by RTPCR method. Results: Compared with the normal control group, the pathological changes of the testis tissue were observed in UU infected rats, the levels of IL1, IL6 and TGFβ were all increased, the expression of Fas decreased and the expression of FasL increased. CONCLUSION: Anti-UU immunoregulation can be exerted in rat Leydig cells by changing the expression pattern of IL1, IL6, TGFβ and Fas, FasL in anti-infective immunity.
