目的观察IgA肾病(IgA nephropathy,IgAN)患者与正常人的血清热聚合IgA1(aggregated IgA1,aIgA1)对足细胞及系膜细胞增殖的影响。方法利用层析法获得IgAN患者与健康人的血清单体IgA1(monom eric IgA1,m IgA1),将m IgA1热聚合为aIgA1,分别刺激小鼠MPC5足细胞及MSC-1097系膜细胞株,利用MTT法检测细胞增殖。结果①不同浓度aIgA1刺激组间足细胞MTT吸光度差异有统计学意义。相比于阴性对照组(SFM),1 mg/m l、2 mg/m l组吸光度分别为0.456(P=0.033)、1.663(P=0.010)。②不同浓度aIgA1刺激组间系膜细胞MTT吸光度差异有统计学意义。相比于阴性对照组(SFM),0.25 mg/m l、0.5 mg/m l、1 mg/m l组吸光度分别为1.667,1.736,1.772(P=0.000),2 mg/m l组吸光度为1.148(P=0.086)。结论足细胞可能会直接对IgA1产生反应;aIgA1可以影响足细胞及系膜细胞增殖,且对两种细胞增殖的影响存在不同。
Objective To investigate the effects of aggregated IgA1 (aIgA1) on the proliferation of podocytes and mesangial cells in patients with IgA nephropathy (IgAN) and normal subjects. Methods Serum monomeric IgA1 (m IgA1) of IgAN patients and healthy individuals were obtained by chromatography, and m IgA1 was heat polymerized to aIgA1 to stimulate mouse MPC5 podocytes and MSC-1097 mesangial cells respectively. MTT assay of cell proliferation. Results ① There was significant difference in the MTT absorbance of podocytes between different concentrations of aIgA1 stimulation group. Compared with the negative control group (SFM), absorbance of 1 mg / ml, 2 mg / ml group were 0.456 (P = 0.033) and 1.663 (P = 0.010) respectively. ② The MTT absorbance of mesangial cells in different concentrations of aIgA1 stimulation group had statistical significance. The absorbance of 0.25 mg / ml, 0.5 mg / ml and 1 mg / ml groups were 1.667, 1.736 and 1.772 respectively (P = 0.000) and the absorbance of the 2 mg / ml group was 1.148 compared to the negative control group (SFM) 0.086). Conclusion Podocytes may directly respond to IgA1; aIgA1 may affect the proliferation of podocytes and mesangial cells, and may have different effects on the proliferation of both cells.
