目的对照研究自发性癫癎大鼠(SER)和正常Wistar大鼠(WTC)脑内Ⅰ、Ⅱ、Ⅲ型钠通道α亚基mRNA表达情况。方法提取枕叶皮质、齿状回及海马CA1和CA3区组织总RNA,应用逆转录-聚合酶链反应(RT-PCR)及建立限制性内切酶图谱检测Ⅰ、ⅡA、ⅡN、ⅢA和ⅢN型钠通道的表达。结果SER枕叶皮质、齿状回及海马CA1和CA3区总钠通道(3.179±1.967、3.009±2.364、3.071±2.036、1.440±0.761)的表达略高于WTC(2.112±1.454、1.474±1.257、2.198±0.978、1.399±0.853),但差异均无统计学意义(P>0.05),SERⅢ型钠通道在海马的表达(1.066±0.276)高于WTC(0.419±0.098),且P<0.01,酶切图谱显示ⅢN型钠通道表达水平有明显上调。结论SER脑内海马Ⅲ型电压门控钠通道N亚型表达上调。
Objective To study the expression of α subunit mRNA of type Ⅰ, Ⅱ and Ⅲ sodium channel in the brain of spontaneous epileptic rats (SER) and normal Wistar rats (WTC). Methods Total RNA was extracted from the cortex, dentate gyrus and hippocampal CA1 and CA3 regions of the occipital lobe. Reverse transcription - polymerase chain reaction (RT - PCR) and restriction enzyme analysis were used to detect the expression of Ⅰ, ⅡA, ⅡN, ⅢA and ⅢN Type sodium channel expression. Results The expression of total sodium channels in SER occipital cortex, dentate gyrus and hippocampal CA1 and CA3 regions was slightly higher than that of WTC (2.112 ± 1.454, 1.474 ± 1.257, 3.179 ± 1.967, 3.009 ± 2.364, 3.071 ± 2.036 and 1.440 ± 0.761 respectively) 2.198 ± 0.978,1.399 ± 0.853), but the difference was not statistically significant (P> 0.05). The expression of SERⅢ-type sodium channels in hippocampus was significantly higher than that in WTC (0.419 ± 0.098, 1.066 ± 0.276, P <0.01) Cut map shows that Ⅲ N-type sodium channel expression was significantly increased. Conclusions The expression of N-type voltage-gated Na + channel in hippocampus of SER brain is up-regulated.
