本文介绍一种新的无血清培养基(Serum free medium,SFM)。这种培养基有利于小鼠骨髓红系祖细胞(BFU-E)生长,这种SFM包含三种不可缺少的基本成分:牛血清白蛋白(BovineSerum Albumin,BSA),胆固醇,和促红细胞生成激素(Erythropoietin.Epo)。且不需另加条件培养基(conditional medium)以补充集落刺激因子(burst-promoting factors)。胆固醇的过滤和BSA的脱脂(delipidation),都可降低该培养基支持BFU-E生长的能力。SEM不仅有利于红细胞系体外培养,且制备简单,经济、高效,培养基中各基本成分均已商品化,容易购买,值得推广。
This article describes a new serum-free medium (SFM). This medium favors the growth of mouse bone marrow erythroid progenitor cells (BFU-E), which contains three indispensable basic components: Bovine Serum Albumin (BSA), cholesterol, and erythropoietic hormone (Erythropoietin.Epo). Without the need for additional conditional medium to replenish burst-promoting factors. Cholesterol filtration and delipidation of BSA reduce the ability of the medium to support BFU-E growth. SEM is not only conducive to the culture of erythrocyte in vitro, but also has the advantages of simple, economical and efficient preparation. The basic components of the culture medium have been commercialized and are easy to be purchased and worth promoting.
