以“富有”和“次郎”的休眠芽为外植体,对甜柿离体快繁技术进行了研究。结果表明,聚乙烯吡咯烷酮(PVP)可有效地防止外植体褐变,提高分化率。芽的增殖和生长适宜的ZT和IAA浓度分别为1~2mg/L和0.05~0.10mg/L。在含有ZT1mg/L的改良MS(KNO3和NH4NO3减半)培养基中加入BA1mg/L,能提高有效新梢率,降低试管苗成本。IAA诱导生根的效果优于IBA,用生长素溶液浸新梢基部后转入1/2MSO培养基中对生根更有利。在较低温度下,以腐殖土作基质,试管苗的移栽成活率可达85%以上。
The dormant buds of “rich” and “Jiro” were used as explants to study the rapid propagation of sweet persimmon in vitro. The results showed that polyvinylpyrrolidone (PVP) could effectively prevent the explants from browning and improve the differentiation rate. The optimal concentrations of ZT and IAA for bud proliferation and growth were 1 ~ 2 mg / L and 0.05 ~ 0.10 mg / L, respectively. The addition of BA1mg / L to the modified MS containing ZT1mg / L (KNO3 and NH4NO3 halved) can increase the effective shoot rate and reduce the cost of plantlets. The rooting effect of IAA was better than that of IBA. It was more beneficial to rooting by soaking the shoot base with auxin solution and transferring it into 1/2 MSO medium. At lower temperature, with humus soil as a substrate, the transplanting survival rate of test tube seedlings can reach more than 85%.
