目的:研究肿瘤坏死因子Ⅰ型受体(tumor necrosis factor receptor type Ⅰ,TNFR-Ⅰ)在大鼠颈动脉体(carotid body,CB)中的表达,以及TNF-α对CB球细胞胞内钙离子浓度([Ca2+]i)的影响。方法:Western Blot和免疫荧光双重染色检测TNFR-Ⅰ在大鼠CB中的表达,用钙成像技术检测TNF-α对大鼠CB球细胞[Ca2+]i的影响。结果:Western Blot结果显示,TNFR-Ⅰ阳性条带出现在55kD处,与其分子量一致。免疫荧光双重染色结果显示,TNFR-Ⅰ强烈表达在大鼠CB球细胞中。钙成像结果显示,外源性给予TNF-α可引起球细胞[Ca2+]i迅速升高。结论:大鼠CB球细胞表达TNFR-Ⅰ,可以感受促炎性细胞因子TNF-α的刺激。
Objective: To investigate the expression of tumor necrosis factor receptor type Ⅰ (TNFR-Ⅰ) in rat carotid body (CB) and the effects of TNF-α on intracellular calcium Concentration ([Ca2 +] i). Methods: The expression of TNFR-Ⅰ in rat CB was detected by Western Blot and immunofluorescence double staining. The effect of TNF-α on [Ca2 +] i in rat CB cells was detected by calcium imaging. Results: The results of Western Blot showed that the TNFR-Ⅰ positive band appeared at 55 kD, consistent with its molecular weight. Immunofluorescence double staining results showed that TNFR-I was strongly expressed in rat CB ball cells. Calcium imaging showed that exogenous administration of TNF-α caused a rapid increase of [Ca2 +] i in the cells. Conclusion: The expression of TNFR-Ⅰ in rat CB cells can be stimulated by pro-inflammatory cytokine TNF-α.
