目的:了解福建省耐多药结核分枝杆菌临床分离株katG基因的突变特点,为建立适合我省快速检测异烟肼耐药方法提供参考。方法:对福建省9个监测点进行耐药性调查,收集菌株经菌种鉴定、药物敏感性试验。选取46株耐多药结核分枝杆菌和15株全敏感株,扩增katG高突变区域的基因片段,测序后比对分析。结果:15株全敏感株未检测氨基酸突变。46株耐多药结核分枝杆菌katG基因的突变率为80.43%,katG 315的突变率为54.35%,联合突变率为52.17%。结论:福建省耐多药结核分枝杆菌杆不同耐药谱katG基因突变率不同,最常见的点突变发生于315位,463位具有等位基因多态性。
Objective: To understand the mutation characteristics of katG gene in clinical isolates of multidrug-resistant Mycobacterium tuberculosis in Fujian Province and to provide a reference for the establishment of a rapid anti isoniazid-resistant assay in our province. Methods: To investigate the drug resistance of nine monitoring points in Fujian Province, and to collect the strains through strain identification and drug sensitivity test. Forty-six strains of multidrug-resistant Mycobacterium tuberculosis and 15 fully susceptible strains were selected to amplify the gene fragment with high katG mutation region, and then sequenced and compared. Results: The 15 fully sensitive strains did not detect amino acid mutations. The mutation rate of katG gene of 46 multidrug-resistant Mycobacterium tuberculosis was 80.43%, the mutation rate of katG 315 was 54.35%, and the combined mutation rate was 52.17%. CONCLUSION: The mutation rate of katG gene in different drug resistance spectrum of multidrug-resistant Mycobacterium tuberculosis in Fujian Province is different. The most common point mutation occurs in 315 loci and 463 loci have allelic polymorphism.
