目的:探讨大鼠视网膜缺血再灌注损伤(retinal ischemia-reperfusion injury,RIRI)过程中,丹参对RIRI的保护作用以及对Bcl-2、caspase-3蛋白表达的影响。方法:随机选取66只Wistar大鼠,分为正常对照组、缺血再灌注组、丹参预处理组,通过对前房加压制作RIRI模型。后两组各分为6、12、24、48、72 h 5个亚组,HE染色观察视网膜组织学变化,SABC法检测Bcl-2、Caspase-3蛋白表达的变化。结果:Bcl-2、Caspase-3蛋白在正常组表达较弱。缺血再灌注组和丹参预处理组6 h开始表达,24 h显著增强,48 h下降,72 h表达减弱,变化趋势基本一致。但是,两组比较,丹参预处理组在不同时间点Bcl-2蛋白的表达均明显强于缺血再灌注组,Caspase-3蛋白表达明显减弱,差异具有统计学意义(P<0.01)。结论:丹参预处理对RIRI可以通过减少神经节细胞的凋亡起到保护作用,其作用机制可能是调节Bcl-2、Caspase-3蛋白表达来实现。
Objective: To investigate the protective effect of Salvia miltiorrhiza Bunge on RIRI and its effect on the expression of Bcl-2 and caspase-3 proteins during the retinal ischemia-reperfusion injury (RIRI) in rats. Methods: Sixty-six Wistar rats were randomly divided into normal control group, ischemia-reperfusion group and Salvia miltiorrhiza preconditioning group. RIRI model was made by pressurizing the anterior chamber. The latter two groups were divided into 5 subgroups at 6, 12, 24, 48 and 72 h respectively. The histological changes of retina were observed by HE staining. The expressions of Bcl-2 and Caspase-3 protein were detected by SABC method. Results: The expression of Bcl-2 and Caspase-3 protein in normal group was weak. The ischemic-reperfusion group and Salvia miltiorrhiza preconditioning group began to express at 6 h, significantly increased at 24 h, decreased at 48 h, and decreased at 72 h. The trend was basically the same. However, the expression of Bcl-2 protein in salvia miltiorrhiza pretreatment group was significantly higher than that in ischemia-reperfusion group at two different time points, and the expression of Caspase-3 protein was significantly decreased (P <0.01). Conclusion: Salvia miltiorrhiza preconditioning can protect RIRI by decreasing the apoptosis of ganglion cells. The mechanism may be that the regulation of Bcl-2 and Caspase-3 protein expression can be achieved.
