目的建立抗菌肽HC-17凝胶剂有效成分的含量测定方法。方法采用高效液相色谱法测定有效成分的含量,以乙腈:0.1%三氟乙酸为流动相进行梯度洗脱,洗脱条件0~25min(23∶77-48∶52),流速1.0mL·min-1,检测波长220nm,柱温35℃。结果凝胶剂的有效成分抗菌肽HC-17在0.20~1.40mg·mL-1浓度范围内线性关系良好(R=0.9995),平均加样回收率为101.47%,RSD=1.35%。结论建立的方法准确、稳定、重现性好,可用于抗菌肽HC-17凝胶剂有效成分的定量分析。
OBJECTIVE To establish a method for the determination of the active ingredient of antimicrobial peptide HC-17 gel. Methods The content of active ingredients was determined by high performance liquid chromatography (HPLC) with gradient elution with acetonitrile: 0.1% trifluoroacetic acid as mobile phase. The elution conditions were 0 ~ 25min (23:77 ~ 48:52), the flow rate was 1.0mL · min -1, detection wavelength 220nm, column temperature 35 ℃. Results The antibacterial peptide HC-17 showed a good linearity (R = 0.9995) in the concentration range of 0.20-1.40 mg · mL-1 with an average recovery of 101.47% and a RSD of 1.35%. Conclusion The established method is accurate, stable and reproducible. It can be used for the quantitative analysis of the active ingredients of the antibacterial peptide HC-17 gel.
