目的观察干扰素(interferon,IFN)对眼眶成纤维细胞(fibroblast,Fb)生成糖胺聚糖(glycosaminoglycansGAG)的作用。方法取5例甲状腺相关眼病(thyroidassociatedophthalmopathy,TAO)患者的眶组织标本进行眶成纤维细胞的培养,另取5例非甲状腺相关眼病患者的标本做对照。在培养的成纤维细胞中加入不同浓度的干扰素-α和干扰素-γ。使用3H标记物和液体闪烁分析器检测干扰素对眶成纤维细胞产生GAG的作用。结果干扰素-γ可明显地刺激成纤维细胞生成GAG且对甲状腺相关眼病患者的作用强于对正常人的作用,干扰素-α无此作用。结论干扰素-γ是眶成纤维细胞产生GAG较强的刺激因子。
Objective To observe the effect of interferon (IFN) on the formation of glycosaminoglycans GAG by orbital fibroblasts (Fb). Methods Five orbital tissue specimens of patients with thyroid associated opthalmopathy (TAO) were cultured in orbital fibroblasts. Five other specimens of non-thyroid associated ophthalmopathy were used as controls. Different concentrations of interferon-α and interferon-γ were added to the cultured fibroblasts. 3H markers and liquid scintillation analyzer were used to detect the effect of interferon on GAG production in orbital fibroblasts. Results Interferon-γ significantly stimulated fibroblasts to produce GAG and had a stronger effect on patients with thyroid-associated ophthalmopathy than normal subjects, with no effect of interferon-α. Conclusion Interferon-γ is a strong stimulator of GAG in orbital fibroblasts.
