目的:分析乳腺癌组织细胞角蛋白5/6(cytokeratin 5/6,CK5/6)表达及其与患者预后的关系。方法:收集北京协和医学院肿瘤医院2000-01-01-2000-12-31手术的乳腺癌石蜡组织标本共307例,制作成乳腺癌组织芯片。采用免疫组织化学MaxVision二步法检测乳腺癌组织CK5/6蛋白的表达,应用SPSS 15.0统计软件分析CK5/6与乳腺癌患者的年龄、月经、妊娠、哺乳、肿瘤大小、TNM分期、ER、PR和HER-2等因素之间的关系及在乳腺癌患者预后中的价值。结果:乳腺癌组织CK5/6阳性表达率为23.9%(68/285)。单因素生存分析显示,CK5/6蛋白的表达与患者的总生存率和无病生存率有关,P值分别为0.022和0.023;将分期、ER、PR、HER-2和CK5/6等指标进行Cox多因素生存分析,显示CK5/6是与总生存相关的独立的预后因素(P=0.023),乳腺癌组织CK5/6蛋白高表达提示有较差的临床预后。结论:CK5/6高表达的乳腺癌患者较低表达患者有较差的预后,CK5/6是与乳腺癌总生存相关的独立预后因素。
Objective: To analyze the expression of cytokeratin 5/6 (CK5 / 6) and its relationship with the prognosis of breast cancer. METHODS: A total of 307 paraffin-embedded specimens of breast cancer from Beijing Union Medical College Cancer Hospital from January 2000 to January 2000 were enrolled in this study. Immunohistochemistry MaxVision two-step method was used to detect the expression of CK5 / 6 protein in breast cancer. SPSS15.0 statistical software was used to analyze the relationship between CK5 / 6 and breast cancer patients such as age, menstruation, pregnancy, breastfeeding, tumor size, TNM staging, And HER-2 and other factors and the prognosis of breast cancer patients in the value. Results: The positive rate of CK5 / 6 in breast cancer was 23.9% (68/285). The univariate survival analysis showed that the expression of CK5 / 6 was related to the overall survival and disease-free survival rate of patients, with P values of 0.022 and 0.023, respectively. The staging, ER, PR, HER-2 and CK5 / 6 Cox multivariate survival analysis showed that CK5 / 6 was an independent prognostic factor related to overall survival (P = 0.023). High expression of CK5 / 6 protein in breast cancer suggested poor clinical outcome. CONCLUSIONS: Patients with low expression of CK5 / 6 breast cancer have a poorer prognosis and CK5 / 6 are independent prognostic factors associated with overall survival of breast cancer.
