基因—放射治疗是近年来国际上肿瘤治疗的一个新策略,但由于实体瘤常处于缺氧状态而对放射敏感性低,影响辐射调控序列的转录活性,因此该疗法目前尚未达到理想效果。在治疗基因上游引入乏氧启动子可克服肿瘤乏氧对辐射调控序列转录活性的影响,提高对肿瘤的杀伤效果。全文对辐射诱导、乏氧诱导以及乏氧/辐射双诱导启动子联合自杀基因放射治疗的研究现状、目前存在问题与前景进行综述。
Gene-radiation therapy is a new strategy in the field of oncology treatment in recent years. However, as the solid tumors are often in hypoxia and have low radiosensitivity and affect the transcriptional activity of the radiation regulatory sequences, the therapy has not yet achieved the desired effect. The introduction of hypoxic promoter upstream of the therapeutic gene can overcome the effect of tumor hypoxia on the transcriptional activity of the radiation regulatory sequence and improve the killing effect on the tumor. The full text of radiation-induced, hypoxic-induced and hypoxia / radiation double-induced promoter combined with suicide gene radiotherapy status quo, the current problems and prospects are reviewed.
