目的研究白细胞介素(IL)-8及其受体(R)A和B mRNA在Hela宫颈癌细胞株的表达及外源性IL-8对Hela细胞增殖的作用。方法实时聚合酶链反应(RT-PCR)方法检测IL-8及IL-8RA和IL-8RB在Hela细胞的表达;MTT法检测不同浓度的外源性IL-8对Hela细胞增殖的影响;RT-PCR方法检测外源性IL-8对IL-8RA和IL-8RB表达的影响。结果 Hela细胞表达IL-8及IL-8RA和IL-8RB,并且IL-8RA的表达量明显高于IL-8RB;外源性IL-8可显著促进Hela细胞的增殖,并明显上调IL-8RA和IL-8RB的表达。结论外源性IL-8可通过上调Hela细胞IL-8RA和IL-8RB的表达而促进其增殖。
Objective To investigate the expression of interleukin-8 (IL-8) and its receptor (R) A and B mRNA in Hela cervical carcinoma cell lines and the effect of exogenous IL-8 on the proliferation of Hela cells. Methods The expression of IL-8, IL-8RA and IL-8RB in Hela cells was detected by real-time polymerase chain reaction (RT-PCR). The proliferation of Hela cells was detected by MTT assay -PCR method was used to detect the effect of exogenous IL-8 on the expression of IL-8RA and IL-8RB. Results Hela cells expressed IL-8, IL-8RA and IL-8RB, and the expression of IL-8RA was significantly higher than that of IL-8RB. Exogenous IL-8 could significantly promote the proliferation of Hela cells and significantly upregulate IL-8RA And IL-8RB expression. Conclusion Exogenous IL-8 can promote the proliferation of Hela cells by up-regulating the expression of IL-8RA and IL-8RB.
