目的:建立HPLC法测定白藜芦醇固体脂质纳米粒(Res-SLN)的载药量与包封率测定方法。方法:采用Topsil C18色谱柱(4.6 mm×150 mm,5μm),流动相为乙腈-甲醇(25∶75),检测波长为306 nm,流速为1 m L/min,柱温为25℃;采用超滤法分离游离药物与白藜芦醇固体脂质纳米粒测定包封率与载药量。结果:白藜芦醇在1.0 1~32.32μg/m L质量浓度范围内与色谱峰面积呈良好线性关系,精密度与回收率均较好;白藜芦醇固体脂质纳米粒平均包封率为81.4%,平均载药量为21.4%。结论:所建方法准确、可靠、方便,可用于白藜芦醇固体脂质纳米粒包封率与载药量的测定。
OBJECTIVE: To establish a HPLC method for the determination of drug loading and entrapment efficiency of res-SLN. METHODS: The mobile phase consisted of acetonitrile-methanol (25:75) with a detection wavelength of 306 nm at a flow rate of 1 mL / min and a column temperature of 25 ° C on a Topsil C18 column (4.6 mm × 150 mm, 5 μm) Determination of entrapment efficiency and drug loading of free drug and resveratrol solid lipid nanoparticles by ultrafiltration. Results: Resveratrol had a good linear relationship with the chromatographic peak area in the concentration range of 1.01 ~ 32.32μg / m L, with good precision and recovery. The average entrapment efficiency of resveratrol solid lipid nanoparticles 81.4%, the average drug load was 21.4%. Conclusion: The established method is accurate, reliable and convenient and can be used to determine the entrapment efficiency and drug loading of resveratrol solid lipid nanoparticles.
