用胶原酶连续灌注大鼠肝脏而获得形态典型、分离率高的肝细胞。用灌注法分离出来的肝细胞成活率高达90%,ATP含量在正常生理范围(8~10n mol/mg干重细胞),肝细胞具有葡萄糖异化作用的酶系能利用丙酮酸、乳酸为底物合成葡萄糖。在MEM培养剂中培养的肝细胞属均匀的单层上皮型细胞,在2~3天内保持原有形态,为非分裂
