应用PCR方法研究了维甲酸(RA)和二甲基亚砜(DMSO)诱导HL-60细胞分化过程中c-myc基因扩增的变化。结果:HL-60细胞内c-myc基因扩增为11,经RA诱导6d后,其扩增程度无明显变化,而DMSO诱导3d后,c-myc基因扩增出现下降,至5d,较未诱导分化前下降约30%。提示了RA和DMSO对HL-60细胞不同的诱导分化机制。
The PCR method was used to study the changes of c-myc gene amplification in the differentiation of HL-60 cells induced by retinoic acid (RA) and dimethyl sulfoxide (DMSO). RESULTS: The amplification of c-myc gene in HL-60 cells was 11. After 6 days of RA, there was no significant change in the amplification of c-myc gene. However, after 3 days of DMSO induction, c-myc gene amplification decreased to 5 days. Decreased by about 30% before induction of differentiation. The different induction mechanisms of differentiation of HL-60 cells by RA and DMSO were suggested.
