Horwell等报道了首例运用培养的绒毛膜绒毛细胞来诊断血红蛋白病。一位27岁的妇女,原籍非州,妊1产0,请求产前诊断镰形细胞病。她和丈夫都知道他们患有镰形性状(HbAs),丈夫的几位侄子有严重的镰形细胞病(Hbss)。病人要求在妊娠前三个月内作出诊断,妊娠16周后,要坚持保胎。妊娠10周,使用1.9mm外径,系有20ml注射器的可伸展不锈钢折管,在超声波引导下,吸取绒毛膜绒毛细胞。首次取得一大块绒毛(约10mg),据认为够作细胞遗传学和生化分析。样品直接收集在培养基,此培养基不含血浆,而含肝素,抗生素,由TC199组成,装在管腔和4 ml注射器内。操作时不需麻醉,病人感觉良好,术后不出血。
Horwell et al. Reported the first use of cultured chorionic villus cells to diagnose hemoglobin disease. A 27-year-old woman, of a non-state origin, has a prenatal diagnosis of sickle cell disease. She and her husband both knew they had sickle-shaped traits (HbAs) and several of their husbands had severe sickle cell disease (Hbss). The patient asks for a diagnosis within three months of pregnancy, and insists on miscarriage after 16 weeks of gestation. Pregnancy 10 weeks, the use of 1.9mm diameter, Department of 20ml syringe stretchable stainless steel folding tube, under the guidance of ultrasound, chorionic villus cells. For the first time a large chunk of villus (about 10 mg) was obtained, which is said to be suitable for cytogenetics and biochemical analyzes. Samples were collected directly in culture medium containing no plasma but containing heparin and antibiotics, consisting of TC199, in a lumen and a 4 ml syringe. No anesthesia during operation, the patient feels good, no bleeding after surgery.
