目的:了解2009年上半年宁波市乙型流感病毒血凝素基因变异情况及种系分布。方法:采用狗肾传代细胞(MDCK)培养分离流感病毒,经血凝和血凝抑制试验鉴定后提取病毒RNA,经聚合酶链式反应扩增目的基因后进行乙型流感病毒的HA1区域基因片段的核苷酸序列测定。结果:2009年上半年宁波市共分离到的21株乙型流感病毒均为V ic-toria系毒株,在5、6月份出现流行高峰。血凝素基因重链区(HA1)基因片段的核苷酸序列测定结果,有两个不同的流行株同时存在,其中新的变异株与B/M alaysia/2506/2004相比已经有6个氨基酸被替代。结论:2009年宁波市乙型流感病毒为V ictoria系毒株,血凝素基因已经发生了一定的改变。
Objective: To understand the variation of hemagglutinin gene and the germline distribution of influenza B virus in Ningbo in the first half of 2009. Methods: The influenza virus was isolated and cultured by MDCK. The virus RNA was extracted after hemagglutination and hemagglutination inhibition test. The HA1 gene fragment of influenza B virus was amplified by polymerase chain reaction Nucleotide sequence determination. Results: The 21 strains of influenza B virus isolated in Ningbo in the first half of 2009 were all V ic-toria strains, with a peak in May and June. There are two different epidemic strains in which the HA1 gene fragments of the hemagglutinin gene are sequenced. There are 6 new mutant strains compared with B / M alaysia / 2506/2004 Amino acids are replaced. Conclusion: In 2009, influenza B virus in Ningbo City was Victoria strain, and the hemagglutinin gene has been changed to some extent.
