目的探讨肝细胞生长因子(HGF)及其受体(c-met)在结直肠癌和非肿瘤性大肠黏膜组织中的表达及其与肿瘤血管生成的关系。方法制作结直肠癌和非肿瘤性结直肠黏膜组织各80例的组织芯片,采用免疫组化技术检测HGF和c-met的表达;以CD105为标记,计数肿瘤组织中微血管密度(MVD)。结果在结直肠癌组织中,HGF高表达者48例,c-met高表达者63例,两者之间有相关性(r=0.231,P<0.05)。HGF和c-met在结直肠癌组织中的高表达率明显高于非肿瘤性结直肠黏膜(χ2=35.387,P<0.05;χ2=59.854,P<0.05)。HGF高表达与结直肠癌有无淋巴结转移及TNM分期有关(χ2=4.743,P<0.05;χ2=5.576,P<0.05);c-met高表达与结直肠癌分化程度和有无淋巴结转移有关(χ2=15.767,P<0.05;χ2=5.765,P<0.05)。在中分化肿瘤中,有淋巴结转移者HGF和c-met高表达明显高于无淋巴结转移者(χ2=13.154,P<0.05;χ2=5.371,P<0.05)。HGF和c-met高表达者MVD明显多于低表达者(t=2.150,P<0.05;t=2.052,P<0.05)。HGF高表达和c-met高表达有相关性(r=0.259,P<0.05)。结论 HGF和c-met的高表达可能与结直肠癌组织的血管生成有关。结直肠癌中HGF和c-met的表达对判断肿瘤恶性程度有一定的价值。
Objective To investigate the expression of hepatocyte growth factor (HGF) and its receptor (c-met) in colorectal cancer and non-tumor colorectal mucosa and its relationship with tumor angiogenesis. Methods 80 cases of colorectal cancer and non-tumor colorectal mucosa tissue were made. The expression of HGF and c-met was detected by immunohistochemistry. The number of microvessel density (MVD) in tumor was counted by CD105. Results In colorectal cancer tissues, there were 48 cases with high expression of HGF and 63 cases with high c-met expression (r = 0.231, P <0.05). The high expression rates of HGF and c-met in colorectal cancer tissues were significantly higher than those in non-tumor colorectal mucosa (χ2 = 35.387, P <0.05; χ2 = 59.854, P <0.05). HGF overexpression was correlated with lymph node metastasis and TNM stage in colorectal cancer (χ2 = 4.743, P <0.05; χ2 = 5.576, P <0.05). High expression of c-met correlated with differentiation and lymph node metastasis (χ2 = 15.767, P <0.05; χ2 = 5.765, P <0.05). In differentiated tumors, the high expression of HGF and c-met in patients with lymph node metastasis was significantly higher than those without lymph node metastasis (χ2 = 13.154, P <0.05; χ2 = 5.371, P <0.05). MVD was significantly higher in HGF and c-met overexpression than in low expression (t = 2.150, P <0.05; t = 2.052, P <0.05). The correlation between HGF overexpression and c-met overexpression (r = 0.259, P <0.05). Conclusion The high expression of HGF and c-met may be related to the angiogenesis of colorectal cancer. Colorectal cancer in the expression of HGF and c-met to determine the degree of malignancy of the tumor has a certain value.
