目的:研究西罗莫司酯化物(Temsirolimus)对肾癌细胞786-0增殖与凋亡的影响,并探讨分子作用机制.方法:采用显微镜形态学观察和MTT法检测细胞增殖抑制;流式细胞仪分析用药48h后肾癌细胞周期变化和细胞凋亡率;Western免疫印迹法检测P70S6K,4EBP-1蛋白水平表达变化.结果:西罗莫司酯化物作用786-0细胞的IC50=3.31μmol/L;给药48h后,细胞明显阻滞于G0/G1期,G2/M期细胞比例减少,细胞凋亡率增加;肾癌786-0细胞中P70S6K和4EBP-1蛋白的磷酸化水平明显下降.结论:西罗莫司酯化物能显著抑制肾癌细胞786-0的细胞增殖作用,阻滞细胞周期于G0/G1期且诱导细胞凋亡,其机制可能与P70S6K和4EBP-1磷酸化相关.
OBJECTIVE: To study the effects of Temsirolimus on the proliferation and apoptosis of human renal cell carcinoma 786-0.METHODS: The cell proliferation was detected by microscopy and MTT assay. Flow cytometry The change of cell cycle and the apoptosis rate of renal cell carcinoma were analyzed 48 h after treatment, and the expression of P70S6K and 4EBP-1 was detected by Western blotting.Results: The IC50 of 786-0 cells treated with sirolimus was 3.31μmol / L; 48h after treatment, the cells were significantly arrested in G0 / G1 phase, G2 / M phase cells decreased, the rate of apoptosis increased; renal cell carcinoma 786-0 P70S6K and 4EBP-1 protein phosphorylation levels were significantly decreased .Conclusion: The compounds of sirolimus can significantly inhibit the cell proliferation of 786-0 cells and arrest the cell cycle at G0 / G1 phase and induce apoptosis, which may be related to the phosphorylation of P70S6K and 4EBP-1 .
