目的:探讨宁神温胆汤对精神分裂症大鼠大脑诱导型一氧化氮合成酶(i NOS)及一氧化氮(NO)蛋白表达的影响。方法:将40只SD雌性大鼠随机分为4组:正常组、模型组、宁神温胆汤组、利培酮组,给予正常组、模型组生理盐水灌胃,宁神温胆汤组灌胃中药药液15.2g生药/kg,利培酮组给予药液灌胃0.18mg/kg。1次/d,连续7d。末次给药后对除正常组外的其他3组大鼠腹腔注射阿扑吗啡2mg/kg制备大鼠分裂症模型,对造模后5、10、15min采用刻板评分方法评定各组刻板行为的改变;15min后断头处死,取脑组织,免疫组化法检测i NOS,硝酸还原法检测NO蛋白表达。结果:模型组刻板评分均在0分以上,表明造模成功;与模型组相比,宁神温胆汤组及利培酮组在5、10、15min刻板行为评分明显减少,差异有统计学意义(P<0.01);与利培酮组比较,宁神温胆汤在5、10min刻板行为减少(P<0.05),而15min刻板行为无差异。模型组i NOS、NO血管蛋白表达均显著高于正常组(P<0.01);与模型组比较,宁神温胆汤组i NOS差异无统计学意义(P>0.05),NO表达降低,差异有统计学意义(P<0.01),利培酮组i NOS和NO血管蛋白表达均明显降低,差异有统计学意义(P<0.01)。结论:宁神温胆汤可以降低大脑组织NO血管蛋白表达,可能是该方对抗阳性精神分裂症作用机制之一。
Objective: To investigate the effects of Ningxian Wendan Decoction on inducible nitric oxide synthase (iNOS) and nitric oxide (NO) protein expression in the brain of schizophrenic rats. Methods: Forty SD female rats were randomly divided into 4 groups: normal group, model group, Nanshen Wendan Decoction group and risperidone group. Normal rats were given normal saline, 15.2g crude drug / kg was given to the Chinese medicine liquid and 0.18mg / kg to the risperidone group. 1 time / d, continuous 7d. After the last administration, apomorphine 2mg / kg was intraperitoneally injected into the other three groups of rats except the normal group to make a rat model of schizophrenia. The stereotypical score was used to evaluate the change of stereotyped behavior at 5, 10 and 15 minutes after modeling After 15min, the rats were killed by decapitation and the brain tissue was taken out. Immunohistochemistry was used to detect the iNOS. Nitric oxide was used to detect the protein expression of NO. Results: The scoring score of the model group was more than 0, which indicated that the model was successful. Compared with the model group, the scores of stereotyped behavior of Ning Shen Wendang Decoction group and risperidone group were significantly decreased at 5, 10, 15 min (P <0.01). Compared with risperidone group, Ningshen Wendan Decoction decreased the stereotyped behavior at 5 and 10 min (P <0.05), while there was no difference at 15 min. Compared with the model group, the expression of iNOS and NO in model group was significantly higher than that in the normal group (P <0.01). Compared with the model group, the iNOS expression in the NanshenWengdan Decoction group was not significantly different (P> 0.05) Statistically significant (P <0.01). The expressions of iNOS and NO in the risperidone group were significantly decreased (P <0.01). Conclusion: Ning Shen Wendan Decoction can reduce the expression of NO vascular protein in brain tissue, which may be one of the mechanisms of anti-positive schizophrenia.
