目的:研究甘木通总黄酮抗心肌缺血作用。方法:取Wistar大鼠36只,随机分为6组,每组6只,假手术组、模型组、甘木通总黄酮低、中、高剂量组(8,16,32 mg·kg-1)和复方丹参片组(250 mg·kg-1),除假手术组外,其余大鼠采用结扎大鼠冠状动脉前降支制备心肌缺血模型,造模成功后ig给药,观察甘木通总黄酮对大鼠心电图ST段位移、血清心肌酶活力及心肌梗死范围的影响;昆明种小鼠30只,随机分为5组,分别为生理盐水组、甘木通总黄酮低、中、高剂量组(12,24,48 mg·kg-1)和复方丹参片组(360 mg·kg-1),采用小鼠密闭缺氧实验,观察甘木通总黄酮对小鼠耐缺氧存活时间的影响。结果:与假手术组比较,模型对照组心电图ST段即刻出现显著性抬高(P<0.01),血清谷草转氨酶(AST),乳酸脱氢酶(LDH),肌酸肌酶同工酶(CK-MB)活力均显著升高(P<0.01),心肌梗死范围显著升高(P<0.01);与模型组比较,甘木通总黄酮各剂量组均可不同程度地降低急性心肌缺血大鼠不同时间点心电图ST段抬高值(P<0.05),降低血清AST,LDH,CK含量(P<0.05,P<0.01),降低心肌梗死率(P<0.01),并呈现一定的剂量效应;还可显著延长小鼠常压耐缺氧时间(P<0.05)。与复方丹参片组比较均无差异。结论:甘木通总黄酮对心肌缺血具有显著的保护作用。
Objective: To study the anti-ischemic effect of total flavonoids from Ganmu tong. Methods: Thirty-six Wistar rats were randomly divided into 6 groups with 6 rats in each group. The rats in sham operation group, model group, low, medium and high dosage of total flavonoids of Ganmu tong (8, 16, 32 mg · kg-1) And compound Danshen Tablets (250 mg · kg-1), except for the sham-operation group, the rats in the other groups were given myocardial ischemia models by ligating the anterior descending branches of coronary artery. After successful modeling, The effects of flavonoids on ST segment displacement, serum myocardial enzyme activity and myocardial infarct size in rats were observed. Thirty Kunming mice were randomly divided into 5 groups: saline group, low, medium and high dose of total flavonoids (12,24,48 mg · kg-1) and Fufang Danshen Tablets (360 mg · kg-1). The mice were subjected to hypoxia test to observe the effect of total flavonoids of Ganmu tong on hypoxia-tolerance survival in mice. Results: Compared with the sham-operation group, the electrocardiogram of the model control group immediately showed significant elevation (P <0.01), serum aspartate aminotransferase (AST), lactate dehydrogenase (LDH), creatine kinase isoenzyme (CK- MB) increased significantly (P <0.01), and the range of myocardial infarction was significantly increased (P <0.01). Compared with the model group, the total flavonoids of Ganmu Tong could reduce the different degree of myocardial ischemia in rats (P <0.05), lower the content of AST, LDH and CK (P <0.05, P <0.01) and reduce the myocardial infarction rate (P <0.01), and also showed dose effect Can significantly prolong the atmospheric hypoxia tolerance time (P <0.05). There was no difference with compound Danshen tablet group. Conclusion: The total flavonoids of Ganmu tong have a significant protective effect on myocardial ischemia.
