本实验选用4~6周龄NC系裸鼠,原代用双侧颈背部皮下接种CNE-2细胞株,鼠间传代用套针双侧接种上代的新鲜瘤块,共传8代,成功率100%。动态观测裸鼠荷瘤后免疫状态变化的结果如下:(1)净体重显著减轻;(2)脾系数增大,瘤重增加,两者间高度正相关(r=0.8395);(3)脾NK活性在第5天时轻度升高,10天时开始下降,15天后显著下降;NK活性与瘤重及脾系数均呈高度负相关(r_1=-0.7132,r_2=-0.8237);(4)第5天时,脾NK细胞体外对人干扰素反应性良好,10天时仍有一定的反应性,15天后失去反应性;(5)荷瘤裸鼠牌细胞自然转化率高于对照组,而对ConA刺激的反应性低于对照组;(6)“癌克星”通过降低血清MDA、提高NK活性及增加瘤组织中单核细胞浸润而起抑瘤生长的作用。揭示在T细胞缺陷的裸鼠,其脾NK细胞是抗移植瘤生长的重要免疫活性细胞。
In this experiment, 4-6-week-old NC mice were selected. Primary CNE-2 cell lines were inoculated subcutaneously on both sides of the back of the neck. Passages between mice were used to inoculate fresh tumor blocks of the previous generation on both sides. A total of 8 passages were transmitted for a total of 100 passages. The success rate was 100. %. The results of the dynamic observation of the changes in the immune status of the nude mice bearing the tumors were as follows: (1) The net weight was significantly reduced; (2) The spleen coefficient was increased, and the tumor weight was increased, and the height was positively correlated (r=0.8395); (3) Spleen NK activity increased slightly on the 5th day, decreased at 10 days, and decreased significantly after 15 days; NK activity was highly negatively correlated with tumor weight and spleen index (r_1=-0.7132, r_2=-0.8237); (4) At 5 days, splenic NK cells responded well to human interferon in vitro, remained reactive at 10 days, and lost reactivity after 15 days; (5) The natural conversion rate of tumor-bearing nude mice was higher than that of the control group, whereas for ConA The reactivity of the stimulation was lower than that of the control group; (6) “Cancer nemesis” plays a role in suppressing tumor growth by lowering serum MDA, increasing NK activity, and increasing mononuclear cell infiltration in tumor tissues. It was revealed that the spleen NK cells in nude mice deficient in T cells are important immunocompetent cells against the growth of xenografts.
