目的 :探讨雌激素对骨的作用机制。方法 :采用体外培养胚胎小鼠长骨模型 ,研究雌激素对骺板软骨细胞核仁组成区嗜银蛋白 (AgNORs)数目和面积的影响。 结果 :当雌酚酮浓度为 10 -7mol/L ,静止区和增殖区AgNORs颗粒面积 /细胞总面积与对照组相比有显著差异 (分别为P <0 0 5和P <0 0 1)。雌酚酮浓度为 10 -8mol/L时 ,只有增殖区与对照组有差异 (P <0 0 5 )。当浓度降至 10 -9mol/L时 ,各区均与对照组无统计学差异。在增殖区 ,AgNORs颗粒数随雌酚酮浓度降低而减小 ,当雌酚酮浓度降至 10 -9mol/L时 ,与对照组无差异。雌酚酮对静止区和肥大区AgNORs颗粒数无明显影响。 结论 :雌激素可能增强软骨细胞的增殖活性 ,促进软骨细胞分裂、增殖 ,从而促进软骨内成骨
Objective: To explore the mechanism of action of estrogen on bone. Methods: The embryonic mouse long bone model was used to study the effect of estrogen on the number and area of AgNORs in the nucleolar organizer area of epiphyseal chondrocytes. Results: When the concentration of estrone was 10 -7 mol / L, the area of AgNORs particle / total cell area in the quiescent zone and the proliferating zone were significantly different from those in the control group (P <0.05 and P <0.01, respectively). When the concentration of estrone was 10 -8 mol / L, only the proliferation zone was different from the control group (P <0.05). When the concentration dropped to 10 -9 mol / L, no significant difference between each area and the control group. In the proliferating zone, the number of AgNORs particles decreased with the decrease of estrone concentration. When the estrone concentration dropped to 10 -9 mol / L, there was no difference with the control group. There was no significant effect of estrone on the number of AgNORs particles in quiescent and hypertrophic areas. Conclusion: Estrogen may enhance the proliferation activity of chondrocytes, promote the division and proliferation of chondrocytes, and promote the formation of endochondral ossification
