目的探讨中国北方辽宁汉族人群HLA区域的MICA和TNF基因微卫星多态性分布。方法采用PCR-毛细管电泳方法检测197名正常无关个体的MICA基因第5外显子和TNF基因复合体TNFa微卫星的基因型。应用Arlequin和SPSS软件进行统计分析。结果辽宁汉族人群MICA基因第5外显子存在5种等位基因,其中以A5.1最为常见(34.01%);TNFa位点存在13种等位基因,其中以a6最为常见(33.76%)。辽宁地区和南方部分地区汉族人群MICA基因第5外显子和TNFa位点的等位基因频率分布均具有统计学差异(P<0.01)。MICA基因第5外显子和TNFa位点间存在一定的连锁不平衡,如单体型A5-a6和A5.1-a7(P<0.05)。结论 MICA基因第5外显子和TNFa位点微卫星多态性分布在中国汉族人群中均存在地区差异。
Objective To investigate the distribution of MICA and TNF gene polymorphisms in the HLA region of Liaoning Han population in northern China. Methods The genotypes of TNFa microsatellite DNA of exon 5 of MICA gene and TNF gene complex in 197 normal unrelated individuals were detected by PCR-capillary electrophoresis. Statistical analysis was performed using Arlequin and SPSS software. Results There were 5 alleles in exon 5 of MICA gene in Liaoning Han population, of which A5.1 was the most common (34.01%), 13 alleles were found in TNFa locus, of which the most common was a6 (33.76%). There were significant differences in allele frequency distribution between exon 5 and TNFa of MICA gene in Liaoning province and parts of southern China (P <0.01). There was some linkage disequilibrium between MICA gene exon 5 and TNFa, such as haplotype A5-a6 and A5.1-a7 (P <0.05). Conclusion There is a regional difference in microsatellite polymorphism distribution of exon 5 and TNFa in MICA gene in Chinese Han population.
