目的:探讨甲基转移酶(DNMT)3a及HLAⅠ类分子在肝癌细胞系中表达及意义。方法:提取HepG2、MHCC97L、MHCC97M3、SMMC7421和BEL7402的mRNA,应用RT-PCR及实时定量PCR检测DNMT及HLA的表达情况。结果:RT-PCR显示,在5个肝癌细胞系中DN-MT1和DNMT3b均高表达。而DNMT3a在高转移的MHCC97M3中的表达低于其他细胞。另外,HLA-A、HLA-B和HLA-C则普遍低表达。定量PCR示,DNMT3a在MHCC97L中的相对表达量为1,在MHCC97M3中为0.33。同时,前者的HLA-A、HLA-B和HLA-C的相对表达量分别为4、4.199和3.9,而在后者中分别是2.129、2.621和2.55。结论:在相同的遗传背景下,DNMT3a和HLAⅠ类分子的高表达与肝癌细胞系的低转移能力相关。
Objective: To investigate the expression and significance of methyltransferase (DNMT) 3a and HLA class I molecules in hepatoma cell lines. Methods: The mRNA of HepG2, MHCC97L, MHCC97M3, SMMC7421 and BEL7402 were extracted, and the expression of DNMT and HLA were detected by RT-PCR and real-time quantitative PCR. RESULTS: RT-PCR showed high expression of DN-MT1 and DNMT3b in 5 hepatoma cell lines. The expression of DNMT3a in highly metastatic MHCC97M3 was lower than other cells. In addition, HLA-A, HLA-B and HLA-C were generally lowly expressed. Quantitative PCR showed that the relative expression of DNMT3a in MHCC97L was 1 and 0.33 in MHCC97M3. At the same time, the relative expression levels of HLA-A, HLA-B, and HLA-C in the former were 4, 4.199, and 3.9, respectively, while in the latter, they were 2.129, 2.621, and 2.55, respectively. CONCLUSIONS: Under the same genetic background, high expression of DNMT3a and HLA class I molecules is associated with low metastatic capacity of hepatoma cell lines.
