应用高效液相色谱法测定食品中的甲醛含量。结果表明:加入0.4 % 的乙酰丙酮溶液4.0 mL,衍生化温度40 ℃,衍生化时间15 min,是最优的衍生条件。实验采用色谱柱为Agilent5TC-C18(2)(250 mm×4.6 mm×5 um),流动相为V( 乙腈):V( 水)=20:80,波长412 nm,采用外标法定量,甲醛质量浓度在1.0 ~ 100 μg/mL 范围内呈良好线性,相关系数为0.9994。选用梨子、大白菜、干墨鱼、熏豆干高中低3 个水平的平均回收率依次为95.1 %、94.7 %、86.4 %、93.6 %,相对标准偏差范rn围为0.93 % ~ 4.28 %。该方法前处理操作简单,环保,结果准确,实验成本低,适合作为测定食品中甲醛的有效方法。“,”A method for the determination of formaldehyde in food by high performance liquid chromatography (HPLC)rnthrough derivatization with acetylacetone was established. The results showed that the derivatization conditions were determinedrnas follows: adding 0.4 % acetylacetone with 4.0 mL at 40 ℃ for 15 min in a water bath.The HPLC method in this experiment wasrnperformed on an Agilent 5 TC-C18(2)(250mm×4.6mm×5um) column by isocratic acetonitril (20%)/water (80%) as mobile phase.rnThe detection wavelength was 412 nm. Formaldehyde was quantified by an external standard method,Under optimal experimentalrnconditions,good linearity was observed in the rang of 1.0~100 μg/mL with a correlation coefficient of 0.9994.The average recoveriesrnof formaldehyde at low,moderate and high spiked levels were 94.6 %、94.9 %、88.8 %、88.8 %,and the relative standardrndeviations(RSD) were between 0.15 % and 1.15 %.The established method is not only simple,environmental and reliable,but also lowrnexperimental cost,which is suitable for the detection of formaldehyde in food.
