目的:观察超微粉化瓜蒌根对2型糖尿病(T2DM)SD大鼠免疫机制异常的干预研究。方法:选用雄性SD大鼠,予高脂高盐饲料喂养8周后加小剂量链脲佐菌素(STZ)注射建立T2DM大鼠模型,大鼠随机分为正常对照组,模型组,中药(低、中、高剂量)组。中药组分别予微粉化瓜蒌根灌胃,正常组和模型组给予等体积生理盐水,治疗4周后采用稳态模式法(HOMA)评价胰岛素抵抗(Homa-IR)和胰岛β细胞分泌功能(Homa-IS)、检测空腹血糖(FPG)及空腹胰岛素(Fins);应用流式细胞仪检测血淋巴细胞亚群分布的变化情况(CD系列)。结果:T2DM大鼠模型呈现CD4+T细胞、CD19+B细胞、CD11a+表达水平显著低下(P<0.05);中药治疗后FPG、Ho-ma-IR显著低于模型组(P<0.05),Homa-IS则显著高于模型组(P<0.05);中药组的CD4+表达显著低于模型组、CD56+、CD14+的表达显著高于模型组(P<0.05)。结论:T2DM大鼠模型存在着特异、非特异免疫细胞功能低下;超微粉化瓜蒌根通过调节淋巴细胞亚群比例分布及降糖、改善胰岛素抵抗和胰岛β细胞分泌功能,对T2DM大鼠具有免疫调节作用。
OBJECTIVE: To observe the intervention of super micronized melon roots on the abnormal immune system in type 2 diabetic rats (T2DM). Methods: Male Sprague Dawley rats were fed with high-fat and high-salt diet for 8 weeks and then given low-dose streptozotocin (STZ) injection to establish T2DM rat model. The rats were randomly divided into normal control group, model group, Low, medium and high dose) group. The rats in the TCM group were intragastrically infused with micronized melon roots, and the rats in the normal group and model group were given equal volume of normal saline. After 4 weeks of treatment, HOMA was used to evaluate the insulin resistance (Homa-IR) and pancreatic β-cell secretory function Homa-IS). The fasting blood glucose (FPG) and fasting insulin (Fins) were detected. The distribution of blood lymphocyte subsets was analyzed by flow cytometry (CD series). Results: The expression of CD4 + T cells, CD19 + B cells and CD11a + in T2DM rats were significantly lower (P <0.05). FPG and Ho-ma-IR after treatment were significantly lower than those in model group -IS was significantly higher than that of model group (P <0.05). The expression of CD4 + in Chinese medicine group was significantly lower than that in model group, while the expression of CD56 + and CD14 + was significantly higher than that in model group (P <0.05). CONCLUSIONS: There is specific and non-specific immune cell dysfunction in T2DM rat model. The superfine melon root can improve insulin resistance and pancreatic β-cell secretory function by regulating the proportion distribution of lymphocyte subsets and hypoglycemic, Immunomodulatory effect.
