目的探讨子痫前期患者内脂素基因启动子-1535C/T多态性与子痫前期发病的关系。方法选择2010年7月至2011年1月福建医科大学附属第二医院收治的96例子痫前期孕妇,其中轻度子痫前期35例和重度子痫前期61例;96例同期正常晚期妊娠孕妇;采用ELISA及Multiplex SNaPshotSNP分型法分别检测血清内脂素水平、内脂素基因启动子-1535位点基因型。结果子痫前期组孕妇血清内脂素水平明显高于对照组,重度子痫前期组高于轻度子痫前期组,差异有统计学意义(P<0.05)。内脂素基因启动子-1535位点基因型和等位基因频率在子痫前期组和对照组中的分布差异有统计学意义(P<0.05)。子痫前期组中,内脂素基因启动子-1535CC型比TT型血清甘油三酯(TG)、内脂素(visfatin)水平明显升高,高密度脂蛋白(HDL-C)明显降低(P<0.01)。结论子痫前期患者内脂素基因启动子-1535CC型与血清内脂素水平、血脂水平相关,参与子痫前期的发病。
Objective To investigate the relationship between the promoter -1535C / T polymorphism of visfatin gene and the pathogenesis of preeclampsia in preeclampsia. Methods Ninety-six pregnant women with preeclampsia were selected from the Second Affiliated Hospital of Fujian Medical University from July 2010 to January 2011, in which 35 were mild preeclampsia and 61 were severe preeclampsia; 96 were pregnant women with normal late pregnancy; Serum visfatin levels and the -1535 locus of the promoter of the visfatin gene were detected by ELISA and multiplex SNaPshotSNP typing. Results The levels of serum visfatin in pregnant women with preeclampsia were significantly higher than those in the control group, and those in the severe preeclampsia group were higher than those in the mild preeclampsia group (P <0.05). The frequency of genotypes and alleles of promoter -1535 of visfatin gene in preeclampsia group and control group was significantly different (P <0.05). In the preeclampsia group, the level of visfatin and serotonin-1535CC was significantly higher than that of TT (P <0.05), and the level of HDL-C was significantly decreased <0.01). Conclusion The PPI-1535CC type in patients with preeclampsia is associated with serum visfatin and lipids, which is involved in the pathogenesis of preeclampsia.
