目的研究大型海藻龙须菜提取物对小鼠脾脏淋巴细胞内TGF-β表达的影响,并初探其免疫调控作用机制。方法以大型海藻龙须菜为实验材料,采用流式细胞术对Balb/c小鼠脾脏淋巴细胞进行表型分析,检测了不同浓度龙须菜提取物共培养条件下CD45+细胞内转化生长因子(TGF-β)表达水平的变化,并对产生TGF-β的细胞进行身份鉴定。结果龙须菜提取物不同反应浓度处理条件下Balb/c小鼠脾脏淋巴细胞中CD45+CD83+/CD45+及CD45+CD80+/CD45+细胞百分率与对照组比较无显著性差异(P>0.05),但CD45+TGF-β/CD45+细胞百分率与龙须菜提取物反应液浓度则呈明显的线性剂量依赖关系(r2=0.8145,P<0.05),最适反应相对百分浓度为2%。TGF-β主要来源于CD3+T细胞(约占35.4%)和CD49b+NK细胞(约占46.6%)。结论提示龙须菜提取物具有通过TGF-β进行免疫抑制或生物调节的功能。
Objective To study the effects of large seaweed extracts on the expression of TGF-β in lymphocytes of mice spleen, and to explore the mechanism of immune regulation. Methods Using large seaweed asparagus as experimental material, the phenotypes of spleen lymphocytes of Balb/c mice were analyzed by flow cytometry, and the CD45+ intracellular transforming growth factors under co-culture conditions with different concentrations of asparagus extracts were tested. Changes in the level of expression of TGF-β) and identification of cells producing TGF-β. Results The percentages of CD45+CD83+/CD45+ and CD45+CD80+/CD45+ cells in spleen lymphocytes of Balb/c mice were not significantly different from those in the control group (P>0.05). The percentage of TGF-β/CD45+ cells and the concentration of the reaction solution of Gracilaria lemaneiformis were linear in a dose-dependent manner (r2=0.8145, P<0.05). The relative percentage concentration of the optimal response was 2%. TGF-β is mainly derived from CD3+ T cells (about 35.4%) and CD49b+ NK cells (about 46.6%). The conclusion suggests that the extract of Gracilaria lemaneiformis has the function of immunosuppression or biological regulation by TGF-β.
