用一噬菌体抗体库分离人杀伤抑制受体特异性的单链抗体。经ELISA分析显示,50个克隆中有14个与NKAT4蛋白反应呈强阳性。这些克隆的重链可变区胚系为DP47。WesternBloting显示表达在上清中的单链抗体为单体。本研究结果表明,从一噬菌体抗体库可比较简便地分离到杀伤抑制受体的特异性单链抗体。
A phage antibody library was used to isolate single-chain antibodies specific for human killer-inhibitory receptors. ELISA analysis showed that 14 out of 50 clones were strongly positive for NKAT4 protein. The heavy chain variable regions of these clones were DP47. Western Blotting showed that the single chain antibody expressed in the supernatant was a monomer. The results of this study show that it is relatively easy to isolate a specific single-chain antibody to a killer-inhibitory receptor from a phage antibody library.
