运用人类高分辨染色体显微切割、PCR和微克隆技术构建的8q24.1带特异性pUC19文库,筛选出一个含CA重复的新的多态微卫星DNA[(CA)n]。经PCR检测人鼠杂种细胞板,证实其来源于人8号染色体;在正常人群中检出11个等位片段,杂合度为0.84;在11个家系中进行连锁分析。结果显示,此多态标记与三个已知的位于8q23-24.1区域的微卫星DNA(D8S85,D8S199,D8S281)紧密连锁,其在8号染色体遗传图谱上的相对位置为:着丝粒-D8S85-新(CA)n-D8S199-长臂末端。
A novel polymorphic microsatellite DNA [(CA) n] containing CA repeats was screened using 8q24.1 band-specific pUC19 library constructed by human high-resolution chromosome microdissection, PCR and microcoloning. The hybridization of human and mouse cell lines was confirmed by PCR, which was confirmed by the human chromosome 8. In the normal population, 11 alleles were detected with a heterozygosity of 0.84. A linkage analysis was performed in 11 pedigrees. The results showed that the polymorphic marker was closely linked to three known microsatellite DNAs (D8S85, D8S199, D8S281) located in the region of 8q23-24.1. The relative positions on the chromosome 8 genetic map were: centromere -D8S85-NEW (CA) n-D8S199-Long Arm Tip.
