目的:建立以高效液相色谱法测定忍冬果实中绿原酸含量的方法。方法:色谱柱为Kromasil C18反相柱(150mm×3.8mm,5μm),流动相为乙腈-0.05%磷酸水溶液=12:88,检测波长为326nm,流速为0.8mL.min-1。结果:峰面积与浓度在30~150μg.mL-1范围内呈良好线性关系,线性回归方程为Y=21370X-303866(r=0.999),平均加样回收率为98.80%,RSD=1.21%(n=9),样品中的绿原酸含量为0.856%。结论:本方法简便、准确、重现性好,可用于忍冬果实中的绿原酸的含量测定。
Objective: To establish a method for the determination of chlorogenic acid in honeysuckle fruit by high performance liquid chromatography. Method: The column was Kromasil C18 reversed-phase column (150mm × 3.8mm, 5μm). The mobile phase was acetonitrile-0.05% phosphoric acid solution = 12: 88, the detection wavelength was 326nm and the flow rate was 0.8mL.min-1. Results: The peak area was linear with the concentration in the range of 30-150 μg.mL-1. The linear regression equation was Y = 21370X-303866 (r = 0.999). The average recoveries were 98.80% and RSD = 1.21% n = 9), the content of chlorogenic acid in the sample is 0.856%. Conclusion: The method is simple, accurate and reproducible. It can be used for determination of chlorogenic acid in honeysuckle fruit.
