目的:研究脉络宁注射液对脂多糖(lipopolysaccharide,LPS)诱导人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)的核转录因子-κB(NF-κB)/组织因子(tissue factor,TF)通路的干预作用。方法:HUVECs在5%CO237℃条件培养于含10%胎牛血清、0.5%双抗和10μg·L-1碱性成纤维细胞生长因子(bFGF)的DMEM低糖培养基中,5 mg·L-1LPS诱导HUVECs 24 h后形成的炎症细胞模型,采用MTT法观察脉络宁注射液对炎症细胞模型活性的影响,从而确定脉络宁治疗组的低、中、高剂量;采用ELISA法检测白介素-6(IL-6)和肿瘤坏死因子α(TNF-α)的活性;采用Western blotting检测NF-κB和TF的含量。结果:与模型对照组比较,脉络宁治疗组的作用增加显著(P<0.05或P<0.01),但在12 mg·L-1后有趋于平稳或下降,则确定脉络宁低、中、高剂量组分别是4,8,12 mg·L-1;脉络宁各剂量组的IL-6和TNF-α的含量都有显著降低(P<0.01)和显著降低炎症细胞模型的NF-κB和TF的蛋白表达(P<0.01)。结论:脉络宁注射液的抗炎作用与其拮抗NF-κB/TF活化有一定的关系。
Objective: To investigate the effect of Mailuoning injection on the expression of nuclear factor-κB (NF-κB) / tissue factor (TF) in human umbilical vein endothelial cells (HUVECs) induced by lipopolysaccharide Pathway of intervention. Results: Compared with the model control group, the effect of Mailuoning treatment group increased significantly (P <0.05 or P <0.01), but after 12 mg · L-1 tended to be stable or decreased, then Mailuoning low, High-dose group were 4,8,12 mg · L-1; Mailuoning dose groups IL-6 and TNF-α levels were significantly lower (P <0.01) and significantly reduce the inflammatory cell model of NF-κB And TF protein expression (P <0.01). Conclusion: Mailuoning injection anti-inflammatory effect and its antagonism of NF-κB / TF activation have a certain relationship.
