利用~3H-TdR掺入率液体闪烁测定法结合台盼蓝拒染率和单层琼脂CFU-GM培养对骨髓造血细胞低温保存后的活力进行了研究。观察了骨髓MNC的不同细胞浓度和孵育时间、不同保存温度(新鲜,4℃,-80℃,液氮)和保存时间(30分,7天,28天,90天)的~3H-TdR掺入年,并与台盼蓝拒染率、单层琼脂CFU-GM培养进行了对比,表明~3H-TdR掺入率是评价低温保存后骨髓造血细胞活力的一项敏感可靠的指标,周期短,并可较准确的反映细胞中处于DAN合成期(S期)的细胞含量。
The viability of bone marrow hematopoietic cells after cryopreservation was studied by liquid scintillation assay combined with 3H-TdR incorporation rate combined with trypan blue exclusion and monolayer agar CFU-GM culture.
