荷叶(lotus leaf,LL)是“药食两用”中药材,具有降脂功效。为观察LL对高脂饮食诱导的非酒精性脂肪肝(NAFLD)的作用及可能的作用机制,该实验采用高脂饲料加饮用果糖溶液建立NAFLD大鼠模型,造模同时灌胃给予荷叶混悬液,血生化仪及ELISA法检测血清ALT,AST,Cr,BUN,TNF-α,IL-6,INS,GLU,ADPN,LEP及肝组织NF-κB,TGF-β1水平;HE染色进行肝组织病理学检查;RT-PCR,Western blot法检测肝组织Adipo R2 m RNA和蛋白表达情况。结果显示,荷叶能显著降低模型大鼠血清ALT,AST,IL-6水平及肝组织NF-κB,TGF-β1水平,升高脂联素水平,增加肝组织Adipo R2蛋白的表达,改善肝细胞脂变。提示,荷叶对高脂高糖致NAFLD具有保护作用,该作用与促进Adipo R2表达,改善胰岛素抵抗,降低炎症因子表达水平,抑制炎症反应有关。
Lotus leaf (lotus leaf, LL) is “Edible and Medicine ” Chinese herbal medicine, with lipid-lowering effect. To observe the effect of LL on NAFLD induced by high-fat diet and its possible mechanism, a rat model of NAFLD was established by using high-fat diet and drinking fructose solution. Serum levels of ALT, AST, Cr, BUN, TNF-α, IL-6, INS, GLU, ADPN, LEP and liver tissue were detected by ELISA, Histopathological examination was performed. Adipo R2 mRNA and protein expression in liver were detected by RT-PCR and Western blot. The results showed that lotus leaf could significantly reduce the levels of serum ALT, AST, IL-6 and the levels of NF-κB and TGF-β1, increase the level of adiponectin, increase the expression of Adipo R2 protein in liver tissue, Cell lipidosis. Tip, lotus leaf on high fat and high glucose-induced NAFLD have a protective effect, and promote the expression of Adipo R2, improve insulin resistance, reduce the expression of inflammatory cytokines, and inhibit the inflammatory response.
