目的探讨白色念珠菌磷脂酶活性与其毒力因子之间的关系以及与其遗传特征的相关性。方法蛋黄平板法检测80株白色念珠菌磷脂酶活性,并测定磷脂酶高活性组、低活性组白色念珠菌的溶血活性、细胞表面疏水性、黏附上皮细胞能力及RAPD-PCR分析2组菌株的基因型特征。结果磷脂酶阳性检出率为97.5%,2组PZ均值分别为0.639±0.043、0.847±0.079(P<0.05);2组的溶血活性、疏水率差异均无显著性(P>0.05),而磷脂酶高活性组菌株对上皮细胞的黏附力显著高于磷脂酶低活性组(P<0.05),磷脂酶活性与黏附力呈正相关(r=0.773,P<0.01)。RAPD结果表明磷脂酶活性与RAPD各电泳条带之间的回归系数不尽相同。结论磷脂酶活性不能直接反应菌株其他毒力的情况,且RAPD分型也不能全面地反映磷脂酶这一单一的毒力特征,而是对菌株毒力特征的一个综合反映。
Objective To investigate the relationship between Candida albicans phospholipase activity and its virulence factors and its genetic characteristics. Methods The activity of phospholipase of Candida albicans was determined by egg yolk plate method. The hemolytic activity, hydrophobicity of cell surface and adhesion of epithelial cells of Candida albicans with high phospholipase activity and low activity group were determined. Genotype characteristics. Results The positive detection rate of phospholipase was 97.5%. The average PZ values of two groups were 0.639 ± 0.043,0.847 ± 0.079 (P <0.05). There was no significant difference in hemolytic activity and hydrophobicity between the two groups (P> 0.05) Adhesion of epithelial cells to phospholipase activity group was significantly higher than that of low phospholipase group (P <0.05). Phospholipase activity was positively correlated with adhesion (r = 0.773, P <0.01). RAPD results showed that the regression coefficients between phospholipase activity and RAPD bands were different. Conclusion Phospholipase activity can not directly reflect the virulence of other strains, and RAPD typing can not fully reflect the single virulence characteristics of phospholipase, but a comprehensive reflection of the virulence characteristics of strains.
