目的检测弥漫大B细胞淋巴瘤(DLBCL)中B7-H1的表达及其对细胞增殖和凋亡的影响。方法通过Western Blot检测DLBCL和瘤周正常淋巴组织中B7-H1的表达水平,将上述2种组织标本进行淋巴细胞分离,利用流式细胞仪对比分析2种组织中B7-H1的表达水平和调节性T细胞(Treg)等肿瘤浸润细胞所占比例。结果 B7-H1在DLBCL组织和瘤周正常淋巴组织中均有表达,但在DLBCL组织中表达更高;B7-H1主要在HLA-DR~+CD14~+巨噬细胞中表达,且在DLBCL组织中表达水平显著高于瘤周正常淋巴组织;在DLBCL组织中浸润的Treg、CD4~+T/CD8~+T比值、p DC要明显高于瘤周正常淋巴组织,上述差异均有统计学意义(P<0.05),而m DC表达差异无统计学意义(P>0.05)。结论与瘤周正常淋巴组织相比,DLBCL中浸润的p DC、Treg及CD4~+T/CD8~+T比值明显增高,提示DLBCL存在免疫抑制,可能是由于B7-H1/PD-1信号通路抑制了T细胞的功能而造成的。
Objective To detect the expression of B7-H1 in diffuse large B cell lymphoma (DLBCL) and its effect on cell proliferation and apoptosis. Methods Western blot was used to detect the expression of B7-H1 in DLBCL and peritumoral normal lymphoid tissues. Lymphocytes were isolated from the above two tissue samples. The expression of B7-H1 in the two tissues was analyzed by flow cytometry T lymphocytes (Treg) and other tumor-infiltrating cells in proportion. Results B7-H1 was expressed both in DLBCL tissues and in normal peritumoral tissues, but higher in DLBCL tissues. B7-H1 was mainly expressed in HLA-DR + CD14 + macrophages, and in DLBCL tissues The expression level of Treg in infiltrating DLBCL tissues was significantly higher than that in normal lymphoid tissues of peripheral lymph nodes, the differences of CD4 ~ + T / CD8 ~ + T ratio were statistically significant (P <0.05), while the expression of m DC had no significant difference (P> 0.05). Conclusions The infiltration of pDC, Treg and CD4 ~ + T / CD8 ~ + T ratio in DLBCL is significantly higher than that in peritumoral normal lymphoid tissue, suggesting that immunosuppression of DLBCL may be due to B7-H1 / PD-1 signaling pathway Inhibit the function of T cells caused.
